On the other hand, the acid digestion method using sulfuric acid (H₂SO₄) and perchloric acid (HClO₄) is more commonly used due to its simplicity and effectiveness, especially for calcareous soils. However, this method may not fully decompose acidic soils, leading to slightly lower results. Phosphorus determination is typically performed using the molybdenum blue colorimetric method. This involves reagents such as ammonium molybdate, potassium antimony tartrate, ascorbic acid, and stannous chloride. Among these, the molybdenum antimony reagent is widely used in China because of its ease of use, stable color development, and resistance to interference from certain ions. The acid-soluble molybdenum antimony colorimetric method involves high-temperature decomposition of soil samples with concentrated sulfuric and perchloric acids to convert all phosphorus into soluble orthophosphate. After filtration, the solution is analyzed using a spectrophotometer at 700 nm. Key instruments include a spectrophotometer, electric furnace, and voltage regulator. Reagents such as concentrated H₂SO₄, HClO₄, molybdenum antimony stock solution, and standard phosphate solutions are essential for accurate measurements. For the alkali fusion method, soil samples are mixed with anhydrous sodium carbonate and melted at high temperatures. The resulting melt is then dissolved in dilute sulfuric acid and filtered. The filtrate is analyzed using the same molybdenum antimony colorimetric technique. This method is more suitable for soils with high levels of insoluble phosphates. Both methods require careful preparation of standard solutions and calibration curves. The working curve is prepared by measuring absorbance values at different concentrations of phosphorus standards. These steps ensure accurate quantification of phosphorus in soil samples, supporting informed decisions in agriculture and environmental management.
